I Hybridoma fusion and screening service: 16 weeks

 

Immunization and anti-sera production

 

8 weeks (USD 1,600)

  • Immunogen requirement:

    • 2 mgs of conjugated peptide (for immunization) and 2 mg of free peptide (for screening).

    • or 3 mgs of purified recombinant protein will be needed.

  • Immunization of 3 mice with standard protocol. Sera from mice will be tested by ELISA using immunogens. Once an acceptable titer is obtained, hybridoma fusion will be done using splenocytes from mouse with the best titer.

Hybridoma Fusion and screening of specific clones

 

8 weeks (USD 3,500)

  • Supernatants from the growing hybridoma wells will be screened by ELISA and western blot using the free Peptide or the purified protein. If the molecular weight and cell type distribution of target protein is known, we will test the ELISA positive hybridoma clones on cell lysate blots.

II. MAP Immunogen monoclonal antibody production

Peptide design and MAP immunogen synthesis

 

3 weeks (USD 600)

  • Custom MAP immunogen synthesis of 15 amino acids. Peptide with more than 15 amino acids will be charged for USD15 per additional residue. We can provide assistance one designing peptide with respect to protein homology, immunogenicity, hydrophilicity, and synthetic suitability.

III. LAE immunogen monoclonal antibody production

Epitope design and LAE Immunogen Preparation

 

4 weeks (USD 1,500)

  • Custom epitope repeat synthesis up to 12 amino acids. We can provide assistance on designing peptide with respect to protein homology, immunogenicity, hydrophilicity, and synthetic suitability.

  • LAE repeat is then reconstructed to link with GST.

  • LAE repeat-GST immunogen is expressed and purified for injection.

  • Epitope monomer is also constructed to an unique PE protein from LAE BIO.

  • Epitope Monomer-PE is expressed and purified for western blotting analysis and other functional studies.

Large-scale culture

 

4 weeks (USD 500)

  • A one-liter culture will be grown from one of the selected clones.

  • Or ascites production in 5 mice for the selected clone.

Purification

 

1 week (USD 400)

  • The secreted antibody will be purified by Protein A affinity chromatography and then lyophilized to send to the customer. Approximate yield may be 20-30 mg.

Antibody Test

 

1 week (USD 400)

  • Immunohistochemistry (IHC) on tissues or cultured cells.

  • Immunoprecipitation (IP) on tissues or cell lysate.
